If mission 128 has a "s129" in it's "on Accept" field,
and if mission 129 requires b0 to be set in order to be available.

Then, if b0 is clear, and you accept mission 128, will 129 be started?

I believe so.

The Sxxx operator AUTOMATICLY starts the mission. Its just overrides the other fields.

Don't want to just be a metoo, but Entarus is correct- Sxxx overrides everything in the availability field. If you want to make 129 start only if b0 is set, you need to make two versions of mďsn 128 and do the availability testing there- if b0 is set, use 128A with the Sxxx in the onAccept. If it isn't, use 128B without the Sxxx.

Or you could always make another mission, requiring b0 to be set, that starts immediatly afterwards: ie the same location.

Keep in mind that while Sxxx will ignore most of the availability requirements of the mission it starts, it does not bypass cargo space needs. Furthermore, there's no way for the first mission to test for this or recover from it in the event that it occurs. For this reason, you should avoid using this operator to start missions with such requirements, unless the first mission has cargo requirements equal to or greater than that of the second one.
